The John Cookson House was built in 1837, and is right on Main Street. John Cookson was the pastor of the First Baptist Church. The house was moved to its current location in 1977 as part of the South End Restoration Project.

The present building was created in 1891, but the original building on site was created in 1835. In 1845 its ownership passed to William Trench, a prominent politician. From 1882 to 1887, it was rented to the Middletown Police and served as their first police station in town.

The Caleb Fuller House was built in 1771. Originally built at the corner of Main and Williams Street by the movie theater and the Inn, it has been moved twice and is now at 49 Main Street. Caleb Fuller was a preacher and eventually became Deacon of the College Church at Dartmouth.

The Middlesex Mutual Assurance Building on Main Street was built in 1867. It housed the Middlesex Mutual Assurance Company, as well as providing meeting space for multiple fraternal organizations to meet. In 1891 an annex was added for a telephone company, and in 1892 a theater was added as well.

The Pythian Building was built between 1873 and 1876 along Main Street. It has held multiple hotels, as well as a Woolworths department store in the 1920s and the Society of Pythias, a fraternal order.

The Caulkins and Post Building was built in 1890, and was considered one of the first modern stores in Middletown. They sold carpets, drapes, and furniture, and by 1903 were also selling automobiles.
